My answer would be to check it out with the website web-developer where you have the problem inputting italics.
ie: A keyboard shortcut may not work on someone's website, unless they allow it's use.

Wondering where it is a problem for you ... ie: where / when do you use italics in Safari ?
Eg: I am using OS Monterey / Safari when I type this, so Command-I works for Italics when typing,
as it does in apps that use text.
Maybe in this case the reply-window's web-developer knew what he was doing.
Seems the input window can (only) accept what it is set-up to accept by the web-developer responsible.
(ie .. useful to ensure correct data, (eg e-mail addresses ... credit card details) in many instances.)
However, when I click Command-I, while browsing a webpage in Safari, Command I goes to the Mail App.
So, it seems the key combination works differently in different contexts:
(Eg: In the Finder, Command-I: shows the Get Info window for a selected file.)
