Mail menu -> Quit Mail.

If the menubar is invisible, hold the escape key to reveal it again.

If it is grayed out, make sure that Window menu -> Activity, has no pending tasks.

If it is grayed out and there are no pending tasks, you can command-option-escape to force quit the Mail application by selecting it, and hitting Force Quit.
