You might have tried typing in a path without the colon after the drive letter to generate that error message, but as you have found even when you use a folder picker to select the target folder it doesn't necessarily stick.
There is a bug in recent builds of iTunes (by which I now mean several years!) that, at least for some users, seems to stop the media folder preference from being saved properly. See Make a split library portable - Apple Community and put your library in a shape that isn't affected by the issue, and is easier to backup and move around in future.

When you change the location of where iTunes files are stored under Edit > Preferences > Advanced, this only affects where new songs and other items that you import are stored. This will not move your entire existing library to the new external drive. This information from Change where iTunes files are stored on PC shows how to move your current iTunes files to the new location using iTunes:
If you want to now move your current files to the new location. see Consolidate your files in the iTunes folder.
If you manually moved your Media folder to this external hard drive already, then you would need to restore your iTunes library so that it will treat the new external drive as the main source of your songs and other items.
First, in the iTunes app on your PC, choose Edit > Preferences, then click Advanced and ensure “Copy files to iTunes Media folder when adding to library” is not checked since this may result in duplicates.
Next, follow these steps to restore your iTunes library but with the Media folder in your external drive as the main source:
This should result in your iTunes Library using your new external drive as the main source for songs and other items.

In my recent post, I assumed that if I typed "Edit>Preferences>Advanced" that sequence would describe the iTunes page I was working with but I see now that I could have been more specific. Let's try this:
Open the iTunes App and click on "Edit". when a menu drops down you will at the bottom the word "Preferences". Click on "Preferences" and a page will open up to show eight columns. The far right column is labeled "Advanced". Click on the "Advanced"gear icon and a page opens up and you will see the line "iTunes Media folder location". The line beneath this line is a line that describes the location of the iTunes Media folder. The description is usually something like "X
x:/Music/iTunes/iTunes Media. The "X" is the drive where the iTunes Media folder resides and that line is the one that must be changed to show the new external drive that you want to use and the pathway to the Media Folder. Let me say here that I am a new member of your group and may not use terms you are used to seeing. Some of you more experienced iTunes experts in the group should comment on the accuracy or lack of it in my post so far.
So now we get to the point where I need some help from the more experienced members of the group. I will repeat part of my earlier post:
I'm having difficulty with the step-by-step sequence that will let me put the new information into the folder at the bottom of the sheet and make it stay there.. The few times I have been able somehow to get the new drive and path put in, after I close iTunes and open it again, and go to the Advanced page I find that the original drive has replaced the new one that I thought I had done correctly. I think the helpful thing you could give me would be to list a step-by-step sequence for me to follow. I seem to be skipping a step somewhere. I'm surprised that the software doesn't ask me to Save the change. If that instruction is present I flat missed it.
I hope this is a good enough description of the problem to allow you to be able to help with it. I will appreciate any help that each of you might be able to give me on this problem. I have to think is that my problem is happening because of some simple step in the sequence that I am skipping. I will thank you all in advance for any help you might be able to give me.

I would like to put my iTunes songs on a different external drive than where they reside now. In Edit>Preferenced>Advanced there is a "Change" button that allows one to designate another drive and path sequence. I just now looked at your post. You have given me exactly what I needed to finally get this thing figured out. Your post contains one of the best step-by-step descriptions I've seen. Also, some info I did not know about that explains why I have been unsuccessful so far. This weekend I will have a chance to implement your instructions and by Monday I will probably be walking around with a big smile on my face.
