Hi! I was asked to provide a permission to a specific folder and occasionally pressed "Cancel"
Can someone please tell me how to set up this permission manually for a specific folder.

Check the "Files and Folders" and possibly the "Full Disk Access" sections of the Privacy & Security System Settings. There should be an entry there which you may be able to manually toggle to "on".

Not every folder on your Mac is owned by you or in your default group, especially those located outside of your home directory and owned by the operating system. Some of these require administrator access and password. You haven't given an example.
I sed folder for downloads for a download manager. Immediately it asked for permission and I occasionally pressed calncel. Now I can't figur out how to make this folder available for the app - it just doesn't as anymore
