Question about turning off iCloud Photos on my iPad 6th gen

If I were to turn off ICloud Photos on my iPad and then delete all the photos off of it, would I have any issues if I were to turn ICloud Photos back on later? Will it delete them from my ICloud backup?

You should not have any issues. Turning iCloud Photos on later would simply re-populate your iPad with the photos.

iCloud Photos has absolutely nothing to do with an iCloud backup. The photos are not contained there.
