This is what I have found as this has happened to me as well. Go to task manager. Right click on itunes.exe then go to priority and set it above normal. This should take care of it. Funny thing is when I spoke with Apple they were of course blaming my computer, connection and anything else and told me a long process to fix it. I explained to them that everything was just fine until the recent update but wouldn't listen. I then found that step after about an hour of searching. Now I have new update (after I got rid of quick time so I have to go back and dowload that and go to the wave.) Personally I absolutely dispise the new Itunes but I think i am past the point of no return. Everytime they update Itunes it skips and all of this and they blame our system when it is what APPLE did.

I saw someone earlier comment on changing quick time default outputs, I'm not sure how the Quicktime settings affect iTunes. But it led me to look at the iTunes settings. Changed Edit / Preference / Playback / Direct Sound / 192kHz / 24 bits sample rate. Restarted iTunes. Skipping gone 100%. Higher quality output to the direct play stereos throughout the house too!

I found that I have to set the priority to realtime. No more skipping. BUT-- I have to change the priority back to Normal (or something besides realtime) before exiting iTunes, or else my computer freezes when I exit. It's a pain in the rear, but at least it works. I can work with other programs when iTunes is open and playing. I haven't noticed any appreciable delays in the other programs in this situation.
I folllowed this advice and it worked. I was running MS Outlook in the back ground. So anything running and syncing in the back ground, may interfere with playback.
We have a winner! I'm using iTunes on Win7 and have tried every idea including inceasing the process priority save reverting to an older release with no success until now. Changing the Quicktime audio preference including "Play Audio Using" safe mode (wavOut only) solved it.

Sorry guys but I've followed these instructions and it's still skipping for me! 😠What version is everyone using? I'm on 10.6.3
Try opening "Audio MIDI Setup.app" and setting all the outputs to 96000Hz 2ch-24bit. This seems to have solved my skipping issue on a Mac Pro. (Audio MIDI Setup is located in /Applications/Utilities/.)

Yep, tried that! Changed the rate, size, channels, and switched to safe mode and it's still skipping.
Just picked up a new iMac (late 2012 model) and, infuriatingly, I am experiencing this same problem. Most of the potential solutions I'm seeing listed concern Macbooks or PCs. Is there an iMac solution that's working for people?
It seems to happen most when I'm browsing the internet while playing music from iTunes LOUD (or at least as loud as the iMac allows). When I turn down the volume, it doesn't seem to happen as much.
Go into Task Manager, right click the iTunes.exe in processes and set it's priority above normal, this should correct the problem.
I'm running Windows 7 Ultimate SP1, I've tried every 'fix' that's been posted... nothing helps. It's not my internet, or my machine, cause everything works in Media Player, and my other music programs. I've kept everything else closed, and stopped all other processes that may be causing it, or took up RAM. I ran a file check, and I also did a security sweep of all file's, folders, and programs. Nothing has fixed it. I'm running it on a Gateway NV52L06u and did not have these problems until the last update. Apple needs to address this issue, as I use it for my guitar covers and I can't have the song skipping every couple of seconds. I have my music on an external, but moved it to the HDD of the computer... same thing. At any rate, they need to fix this. Completely unacceptable from Apple.
1.Download Ccleaner. 2. Put anti-virus/firewall etc... on hold or "silent mode," 3. Run CCleaner 4. switch to Tools-Startup-select all under each tab and turn to disable. 5. Restart computer. Fin.
Control Panel / Hardware and Sound then under Sound, manage audio devices.
Playback, properties, then advanced.
Uncheck 'allow applications to take exclusive control of this device'.... I also put the bitrate to max.
No more skipping.
I found a solution, too.
New computer, new version of iTunes. No skipping. Seriously. I had tried everything else still skipped. I concluded that my computer was old as dirt and needed to be replaced. 🙂 It needed to be replaced for other reasons. But I'm thrilled that I can actually use my iTunes without the annoying skipping.
I experienced the problem since iTunes' September update. Rebuilding the iTunes Library did the trick for me:
