Presuming you're running iTunes 10.x, or at least iTunes 9.2,1, problems connecting to the iTunes Store often stem from issues with the Windows firewall or a firewall in a router, a privacy filter (such as McAfee Internet Privacy Service or Norton Internet Security), an antivirus/antispyware utility, or a download accelerator. for some basic tips to start isolating the cause of the problem. What you may need to do is temporarily disable any such service and see if that fixes the problem. If so, then if it's a firewall/Internet filter that's causing the problem and you'll need to set the offending service to allow iTunes to connect to the iTunes Store.

I had the same problem. And firewall is not the cause of it. What you need to do is :
> start menu
> accessories,
> right click on the command prompt icon and choose "run as administrator".
Once it opens, type in the following command...
netsh winsock reset
hit enter
You should get a message that the winsock reset successful and you will need to reboot your computer.
Reboot and when I reloaded itunes the store loaded fine.
It worked for me. I have windows vista.

I didn't get the prompt on a right click with accessories...

so frustrated. This didn't help us. I will keep looking and keep trying. We have used iTunes for years, but the last few months we jhust can't get on at all.
The "Winsock update" didn't work for me either. I've ran all the diagnostic checks on itunes and all checks come out as passed. When I go to update, it makes it as far as backing up, (backing up to computer, not I Cloud) and it freezes. (See screen shot)This is only slightly better than this one. Both taken AFTER reinstalling and running the "Winsock update".This is effing annoying to say the least !!! Any suggestions ? It seems that as soon as Itunes is launched, my computers performance goes to crap. Task manager shows that as soon as Itunes is launched, memory usage goes up to 100%. I can view the apps update, and that shows up fine. However, I can't get to the store at all on my computer. My iphone connects just fine though.

Once you identify the LSP, google the offending software and you should identify the programme doing the blocking.
My culprit was my Vodafone Mobile connect.
Deleted this, restarted, itunes stores opens, reloaded latest Vodafone software, everything works.

Nothing worked for me either LSP thing was fine netsock didn't work...spent a gazillion hours looking at all the articles. Then I tried changing my default browser from Chrome to IE and bingo..worked perfectly, just restarted itunes.

I can do the winsock reset and the iTunes Store works, but it fouls up the content filtering application on my PC, Safe Eyes. This is the response I got from their tech support when I inquired:
We have identified areas of conflict between Apple's latest version of iTunes, and third party content filters and security applications, including SafeEyes. Currently, our development team is researching and testing the best ways to resolve these conflicts. At the moment, there is no time-line available as to when a patch will be available, but we do expect it to be soon. I apologize for the inconvenience and for your patience.
I have the same type of program "Cloud Care" on my computer, with the same issue. Pretty lame !!!
