Go to apple website and leave a feedback their. people should report this bug through feedback directly to apple. I already have done it.
I suppose Apple has responded by firing Scott Forstall. 😝
I have the same issue. But I haven't been using the Assistive feature. It's been off the whole tim.
I also have this problem and do not use assistive touch. Auto lock was working fine til I upgraded my iPad to 6.0.1 yesterday.
This CANNOT be a "new feature" to save you the hassle of having to unlock your phone every time you need it cos I'd rather do that than take my phone out of my pocket 5 hours after I last used it to find that the battery is down to 30%.
What if, after an 8 hour drive you break down and need to phone to be rescued so you take your phone out and, to save you the hassle of having to unlock the phone, the battery ha completely run out an your stranded??
This is a bug. Temporary solution until apple fix it:
1. Don't use Assistive Touch at all.
2. IF you need to use it, after press teh Assistive Touch home button, touch or tap anywhere in the screen, it should start countdown to Auto-lock.
The second solution works great, altough I may forget to tap the screen after I hit home, at times. 🙂
IOS 6.1 seems to solve both auto lock and camera problems in my 4s. Finally it worked without any trouble eventhough using the assistive touch. One remarkable other thing is that my phone is faster than before. Thats of course very nice! I love to use my iphone more than before! Thanks Apple !!!
I have an NEW iPhone 5. I was JUST at the apple store last night with this problem. I am NOT using Assistive Touch, nor did I know it exsited!
THIS IS A BUG! The apple tech's solution was to just set me lock screen to off! I AM pretty ticked off now! My screen just stays on! I guess I will be bach to tech support!
APPLE, if you know this is an issue in some units, FIX IT OR GIVE ME ANOTHER PHONE!!!!!! Says a LOT!

Somehow the rest of the 100 million iP5 ownners have manged to escape your "bug", which has absolutely nothing to do with the one discussed in this thread.

Issue Resolved in IOS 6.1 update.
i updated my 4S yesterday to IOS 6.1 and up till now the autolock is working fine while using the AssistiveTouch option.
so update ot IOS 6.1 and the issue will vanish.

same here...4S with assistive touch enabled...
but even if usually i use sleep/standby button, batt drain still....sigh...
we need fix from 6.1.2!!!
Its a possibility that your phone isnt locking because a music playing app is running in the background. My iPhone 4S wasnt locking automatically and after trying the method of swiping the screen after using assistive touch (with no success) I closed my music app and that did the trick even without swiping the screen after using assistive touch.
