When on the import menu, the Desktop tab on the left hand side has a little buffer symbol next to it and I can't click on it or access the files to import them.
This also happens on iMovie. Does it have something to do with my hard-drive?

Run Repair Permissions in Disk Utility.

I ran Repair Disk Permissions, it completed. Do I need to restart my system or anything? Tryed to import a file but its still a problem.

I had to go out for a few hours. Is the Desktop/home folder problem still unresolved?

Unresolved still. Called up Apple and they suggested trying to re-installing OS X. Done that, but am still having the same issue.
Right, just made a second user and logged in to it and it seemed to work fine.
So is the best thing to do now, transfer my files into the Shared folder, make a new account and then delete my current one?

Is your home folder (Oli) now located where it's supposed to be?
I was curious about whether your Oli folder was somehow hidden in another folder – or whether there is another folder with the same name somewhere. It would be interesting to do a Spotlight search on Oli.

I still can't access the home folder from Final Cut Pro or iMovie. I've been using the second account I made for the time being to do my editing on.
On my Oli account, I searched 'Oli' in the spotlight search, then clicked 'Show All in Finder'.
Then in the finder that popped up, under the header of 'Folders', it only showed the new user I had made, 'Oliver'.
On the Finder, I then selected my mac under Devices, and in the User folder, it only had 'Oliver' (the new user account) and 'Shared'.
I think I will just have to setup the new user and delete the 'Oli' one.
