I have the same problem. I found a different work around. I went to my local NPR station channel (KUT) and the AirPlay icon was there.
Not that this fixes the real issue but it is one more way to get around the issue. Of course, it only works if the program you want to hear is actually also being simulcast on your local station.

Well, so much for that theory. The AirPlay icon is there when I start streaming but then goes away.

Work around:
File>Open Stream (Command + U)
enter streaming address e.g. http://www.wnyc.org/stream/wnyc-am820/aac.pls
Airplay worked with the old internet radio links and it works with individual stream links, but it doesn't seem to work with talk radio stations on iTunes Radio. Annoying, but at least there's a work around for your favorite stations.
Running Yosemite w iTunes 12 and seeing same problem. No airplay with NPR. I do not understand how to locate the url... this info seems hidden. I did locate a url of a random station and entered it and got the airplay icon... but would not play out the selected devices. This is a real bummer.
I found another work around. First you must have your sound icon visible in the menu bar of your desktop:
Next get your NPR running on iTunes. Initially you will have to run it out of your default speaker. Once NPR is streaming click on the sound icon while holding "option" You should be get something like this:
from there you can select other airplay devices. The only problem is that you cannot simulateously stream to multiple speakers.

Just adding myself to the list of people who are experiencing this issue. I was actually able to stream NPR stations just fine over AirPlay until my last iTunes update (or maybe two updates ago. Currently I'm on iTunes version 12.1.0.50 and OS 10.10.2 on a 2012 MacBook Pro). Very annoying, because I've used iTunes to listen to Radio for years. Now that video commercials show up whenever I start streaming my NPR station, it's probably time to start looking for alternatives.
I'm running iTunes 12.1.2.27 on a Mac Pro/Yosemite.
Suddenly, after working perfectly forever, radio streaming drops out after about 5 minutes. This happens through AirPlay to a stereo system or locally through the computer's speaker system.
The streaming will continue playing only if I use the stream available through the Radio tab in iTunes. Unfortunately, the AirPlay button disappears and is completely unavailable when streamed in this fashion.
PLEASE: HOW TO FIX!??!
Sadly, it's 2016 and this is still a bug on my iMac with 21.5 inch Retina display running Sierra and iTunes 12.5.3.16. Watching the GUI refresh, I can see that iTunes is drawing the icon for choosing Airplay but then drawing the mini-icon for the radio station on top of it. Unfortunately, the work-arounds listed don't work for me.
I can use my iPad, but streaming from my iMac (normally) lets me stream to two separate stereos (plus my computer), while the iPad is limited to only streaming to one device.
Hi, Looks like this is an old thread but I'm having the exact same problem. Open stream (Command-U) work-around does work but not ideal. Sierra on MacBook Pro, iTunes 12.7.0.
