Nope. Various models of iMac can be used as a display and can use external displays (as was mentioned), and screen sharing and remote access is also an option (as was mentioned), and there are various ways to share files and directories among both devices (including iCloud), but the MacBook Pro internals cannot be bypassed and that device then used as a keyboard and mouse for the iMac.

Your post title indicates that you want to use screen sharing to have you MBP access the iMac remotely. If this is the case, then is should be a simple matter of doing, as a minimum, the following two things:

1. On the iMac, go to Settings > Sharing > Screen Sharing, and enable it.
2. On the MBP, open a new Finder window, select the iMac, and then, select Screen Sharing.

And if your iMac is new enough (Late 2009 iMac) you might be able to use Target Display mode as indicated by Eric Root

Clarification question - are you wanting to use the iMac only as a monitor or are you trying to use the iMac's computer system?
