How to prevent waking up and access the external hdd (set for Time Machine (manually)) during the sleep mode.

Not a good idea since your Time Machine backup drive may well disconnect after going to sleep. No backups will occur after wake-up because the backup drive is no longer connected. I will have been improperly ejected.
Point is that TM Backup set to off (manually) and I don't need to macos to access the external hdd during the sleep mode.
I can start TM Backup later manually when I needed.
I need to prevent system to access the hdd during the sleep.

The point should be what other backup utility should you be using? Time Machine isn't it given what you have just stated. Why do you need to prevent the system from accessing the backup drive during sleep? Time Machine is intended for use as a regular cumulative backup every hour. There are other backup utilities with more flexibility in scheduling.
Main Question is what the reason to access the external hdd during the sleep especially when the auto backup is turned off? What system trying to do on that disk – I need to keep disk off and quite and prevent access unnecessarily.
I use TM in manual mode and turn on only during the heavy load work – no need to backup every hour – prevs backup already catch enough information...

Yes, that checked...
I think we should suspect something like logging or so... investigate what system do when access that disk...
Logging was turn off via terminal (trying to find out exact terminal command I apply...)

What is the real rationale for all this effort to completely cease any disk activity? Why not just find a utility or Unix command that parks the drive's head (won't work for an SSD.)
You are right I have to keep the external disk untouchable unless I needed...
