I have created a teacher resource for sharing as a PDF digital file. On the document I have added a media audio file. However when printed or saved, the audio button is then not visable and therefore has no placeholder to show its there or to select. to play.

You cannot insert audio or video from Pages into an exported PDF. You would need a purchased PDF Editor to further this goal, and Apple's Preview is not that tool. The inserted audio, or video object is only supported in the Pages document itself.
