While still in the project, you can make changes to a previously set Picture in Picture by selecting the overlay clip and opening the Picture in Picture feature again. Also, you can select the clip and click on the "Reset All" item, written in white, in the upper right hand corner of the screen. That will erase all settings previously made with Picture in Picture and you can start from scratch.
Once the movie has been shared out to a final product you cannot make changes to the Picture in Picture. You would need to go back to the editable project itself, make the changes, and then share out again.

Hi, fisher63,
Could you provide more details? Which of the overlay options are you using -- cutaway, green screen, split screen, Picture in Picture? What type of changes to you want to make?
Generally, the controls for each option appear just below the tool bar in the upper right hand corner of your screen, when the option is selected. With Picture in Picture you can work in the Preview screen to drag the position of the picture where you want it, and drag the corners of the picture to adjust its size.

Rich: I understand how to utilise all the different functions for the picture in a picture but want to know how to make subsequent changes after the initial screen has been created and the file rendered. Is this possible?
Thanks,
Gary
