Is anyone experiencing an issue where after the latest iOS upgrade all mail that comes into your inbox appears read before you open the mail? If so, what is the fix?

Not an issue that I have seen reported. Have you updated to iPadOS 14.6 - released yesterday? Of not, try updating to this latest version.
If you have already updated, a forced-restart of the iPad may be helpful - as doing so will often resolve an isolated software issue:
- On an iPad with a Home button: Press and hold the top button and the Home button at the same time. When the Apple logo appears, release both buttons.
- On other iPad models: Press and quickly release the volume up button, press and quickly release the volume down button, then press and hold the top button. When the Apple logo appears, release the button.
