I have the exact same issue, Safari worked at first after I got Yosemite, but now I can't launch the app at all. The icon just bounces once and stops. Help!
EDIT: I've also just noticed that Mail doesn't want to launch either. Great.
I have the same problem, also Mail, App store, and Terminal stop working, they all bounce just a once and nothing open.
however iTunes works great.
hope apple resolve this thread as soon as possible.

after reinstalling Yosemity my macbook is working fine.
no problem any more with safari Or any app.
what i did is turning on and pressing COMMAND +R.
giges the option of reinstalling the software.
does not erase the hard drive. All your info documents stay there.
takes about an hour to complete but it fixes the issues.

I have the same problem and it's driving me crazy. Keep regretting my decision to upgrade.. I should have stayed still and ignored the announcement.
Same problem here. So annoying!!!! Please Apple do something. Safari, e-mail, App store not working with Yosemite update. They worked for a little while and then stopped. Can someone from Apple read this and respond?
Plus I don't like how it looks, my son asked my why the computer screen looks older. It seems Apple wants to look more like an Android or Windows product. Its missing the nice clean graphics they used to have, the same happened with the iPhone upgrades.

I had the same trouble
option + command + R
While rebooting will start your computer from internet, from there you can choose, among other options to reinstall Yosemite (wich solved my problem)

I tried that but says I can't do that because I "have no internet connection." Which I do. Because I'm typing this in Chrome.
I have found a temporary solution that works until reboot.
Open a Terminal window and type:
sudo launchctl unload -w /System/Library/LaunchDaemons/com.apple.discoveryd.plist
-type password-
then:
sudo launchctl load -w /System/Library/LaunchDaemons/com.apple.discoveryd.plist

I have the same problem, only worse - Safari's gone, and so is the app store. When I try to update software or download any software anywhere, the computer freezes up.
And I can't reboot from the original disk - this is an update that came from online. And I don't have the reboot disk which would get me back to un-horrible.
Has Apple destroyed my computer with this lousy product?
I have a similar Safari problem. I've had three Apple tech support conversations so far stretching over the last 6 days. I have reloaded Yosemite (5 hours) tweaked preferences, verified disk, etc. No luck. No problems were found (?) It's worse than ever. Mail doesn't work either. Chrome is fine. The Apple telephone support people told me I have to take the computer to an Apple store, which I will do next week. Has anyone found a definite fix?

Actually, my problem with the new yosemite was my mail, app store, safari, terminal, system information all stops working after I downloaded javaforos2014 for my mac. The java is needed for my adobe product to work, so i can't help it but to download it. After installing yosemite everything was working fine until the point that i have to use my flash which promt me to download java, without hesitation i quickly downloaded it and starts to realize that those programs that i mentioned earlier stops working. Following some advice i rebooted and reinstalled yosemite (yes, it took me another 5 hours to download and install) again and everything is working untill i downloaded java again...so that's when i realized that if i want to continue using my adobe's product i have to let go something i.e things that i've mentioned earlier (~_~;)
I had the same problem. After install everything worked fine. After reboot, native apps will not launch.
The command + option + r sequence helped. Time consuming. Now it all works. Hopefully the problem has been fixed.
Apple really needs to look into this pesky problem
