When clicking on the mp3 music file either from desktop or the music folder it plays fine in iTunes. It just doesn't show anywhere.
In that case, it is listed in your iTunes Library, somewhere.
iTunes is simply a database, (a list). The music is not "in" iTunes. Instead, it is in the folder that you already know of and all iTunes does is keep a note of where that is and then refer to that location when you ask it to play or be added to an iPod or iPhone (yes, okay, or iPad)
Did you do as I suggested and switch to the Songs view, then look for the Date Added (to Library)? Mind you of course, if you were looking at a more recent date (the one when you think you re-added the song, rather than the original add date, you may not have seen the entry.)
Try this instead:
- In your music folder, double-click the song so that it begins playing
- Once iTunes is playing the song, it will appear in the Now Playing panel at the top of the iTunes window

- In the screenshot above, notice that the song "Balearic Breakfast" is playing, but the song "Life's A Long Song" is highlighted in the song list panel below it
- If you hover your pointer (mouse) over the Now Playing panel, three blue dots will appear to the right of the song title

You can left-click or right-click on the three dots (make sure it is on the dots) and the following context menu appears:

Notice in the screenshot above, that the song Life's A Long Song is highlighted in the song list panel. If you select Go to the Current Song from the context menu, the song list panel will now go to and highlight the currently playing song

- Notice the speaker icon to the left of the song title in the song list panel
- This entry is the iTunes entry for that song. But if things such as the song title are missing, it makes it much more difficult to find the song when required
- If some details are missing, add them in now by using Edit/Song Info
